The increase in longevity is associated with a wider prevalence of heart failure with preserved ejection fraction (HFpEF). While the incidence of HFpEF in terms of mortality is constantly growing, treatments remain unsuccessful because of insufficient knowledge on the mechanisms of cardiac ageing. In an original approach, our consortium proposes to develop and share appropriate models to better understand HF in the context of ageing.

Role of Type IV pilus retraction in bacterial pathogenesis – RETRACTOPATH
Type IV pili (Tfp) are highly dynamic long filamentous structures that are produced by numerous pathogenic bacteria. They mediate an extraordinary array of functions, including motility, DNA uptake, bacterial aggregation or adherence to host cells. Pilus retraction generates remarkably high force. Such force acts as a mechanical signal to both bacteria and host cells. To date how such force is sensed in host cells and contribute to bacterial pathogenesis has been poorly explored.
Role of Tbx3 in directing the functional identity of POMC neurons in obesity – neuroIDobese
Chronic consumption of hypercaloric diets may trigger a ‘neuronal identity crisis’ in certain subpopulations of hypothalamic neurons due to the impaired action of the T-box gene 3 (Tbx3), a transcription factor that maintains hypothalamic neurons identity during postnatal life. Such a loss of identity may undermine the working machinery of POMC neurons, ultimately leading to obesity and its associated metabolic complications, albeit this hypothesis has yet to be investigated.
Role of Piezo1/KCNN4 interaction in red blood cell physiopathology – ROPKIP
Activation of the mechanosensitive channel PIEZO1 in the red blood cell (RBC) membrane induces a calcium influx responsible for the activation of a KCNN4 potassium channel. The functional coupling between these two ion channels leads to a leak of K, Cl and water and therefore dehydration of the RBCs. Independent mutations on either of these ion channels lead to erythrocyte dehydration responsible for the rare hemolytic anemia, hereditary dehydrated stomatocytosis.

In pulmonary hypertension (PH), both an increase in arterial reactivity and a structural remodeling, causing a thickening of the muscular medial layer of small pulmonary arteries (PAs), are implicated in the etiology of the disease.Several arguments point to an important role for mechanical stress in the stimulation of PA smooth muscle cells (SMCs) proliferation. We hypothesize that opening of the mechanosensitive ion channel Piezo1 in PA SMCs is implicated in experimental and human PAH.

Ocular rosacea (OR) is a common but underdiagnosed ocular surface disease. It alters significantly the quality of vision and life and can threaten the visual prognosis as no treatment is available. OR results from chronic neurovascular and inflammatory mechanisms that can be triggered by corticosteroids. But the mechanisms linking corticoids and neurovascular, immune dysregulation and meibomian gland dysfunction (MGD) in OR are unknown.
