Contrôle stéréochimique de l’auto-organisation supramoléculaire d’analogues de lipides membranaires d’archaea – StereoArchaea

Submission summary

The cell membrane of archaea are composed of very specific membrane lipids that are responsible for a fine tuned modulation of the cell membrane properties (rigidity, fluidity, permeability, asymmetry of the membrane,..). The specific structures of these lipids are directly related to these properties, however structure/property relationships are not clearly established and even less about stereochemical effects. A better understanding of this relationship would permit to enlarge the scope of application of these lipids or of their analogues. This project aims to determine the relationship between the stereochemistry of archaeal lipid analogues and their self-assembling properties. Thus, the project will consist in the synthesis of a series of stereochemically pure archaeal lipid analogues including the main features of natural archaeal lipids and bearing adequate polar head groups to reach a suitable lipophile/hydrophile balance. Recording physico-chemical properties of the resulting self-assemblies would then permit to establish relationships between the stereochemistry of the lipids and their self-assemblies. The properties of the membrane involved in the corresponding objects or Langmuir-Blodgett monolayers are expected to be affected by the variation of some stereochemical centres on the lipid structure. Particularly unsymmetrical membranes would be reachable through a fine control of the molecular symmetry of the tetraether lipids. This project would then result in a better understanding of the cell membrane properties of these particular archaea organisms and it would permit the development of new lipids capable of unique self-assembling properties (enhanced membrane stability, low permeability, controlled membrane symmetry, formation of chiral nano-objects). Therefore this project wants to be the foundations of more ambitious programs and due to these specific properties it would initiate the development of applications in nanotechnology that require precisely these type of membrane properties. For example in the following two areas 1) The study of artificial but functional archaea cell membranes (proteoliposomes) and 2) The use of these lipids in nanotechnology applications (sensors, organic nanotubes, ..).
