Experimental Constraints on the Origin of Volatile Elements on Earth – VOLTERRE
Volatile elements (e.g. H, C, S) have a fundamental role in planetary evolution. The goal of the proposed research is to better constrain how and when Earth acquired its volatile elements. An original and complementary multi-techniques approach combining experiments at high pressure and high temperature, stable isotopes geochemistry, quantitative analyses at high resolution and synchrotron techniques will be developed to tackle the issue of volatile accretion history of the Earth.

In previous reports, we have shown that small molecule inhibitors of EthR stimulate the transcription of the ethA gene, which improves the bio-activation of ETH and consequently boosts its antibiotic activity, both in vitro and in vivo. However, as expected, these molecules were unable to boost the bio-activation of ETH in strains harboring mutations in ethA. Tea-4-Two is based on the discovery of a new generation of drug molecules that main circumvent the problem of drug resistance to ETH.

Obtaining reliable estimates of mutation fitness effects and characterizing directly the dynamics of mutation appearance was impossible previously since mutations have never been witnessed in action in single cells. Such estimates have wide application and are critical for most evolutionary studies. We accomplished this in Escherichia coli by employing microfluidics, time-lapse imaging, and by visualizing mutations in single cells by using a fluorescent tag of the Mismatch Repair System.
